New Reagents for Isolating DNA

A complete and ready-to-use group of reagents is designed for the isolation of genomic DNA from blood, cells, and tissues. The isolation procedure using these reagents is completed in 20-35 minutes.

The DNAzol reagents from Invitrogen (Carlsbad, CA, USA) are used for isolating DNA that is to be used for the diagnosis of a wide range of diseases. The DNAzol procedure is based on the use of a novel guanidine detergent lysing solution that hydrolyzes RNA and promotes the selective precipitation of DNA from the cell lysate. The reagent is not toxic to cells and the procedure does not require phenols.

A biologic sample is homogenized and lysed in DNAzol at room temperature. The sample can be stored or used immediately. The DNA is precipitated with ethanol, washed, and dissolved in 8 mM sodium hydroxide (NaOH). After pH adjustment, the samples are analyzed or stored at 4?C. A wide range of DNA molecules can be isolated, including fragments as small as 100 bp (base pairs) long. In general, 70-100% of DNA is recovered using this procedure.

The isolated DNA can be used for Southern blot analysis, dot blot hybridization, molecular cloning, and polymerase chain reaction (PCR,) without additional purification. There are three types of DNAzol that can be used for isolating blood from three specific types of samples. The DNA extracted is frequently used in PCR and RAPD (random amplification of polymorphic DNA) tests for the diagnosis of fungal disease.
