Assay for Fluorescence Detection of APC Resistance

A simple, low-cost assay has been developed to detect activated protein C (APC) resistance in blood plasma using fluorescence methods.

The new assay provides data in concordance with the existing method of detecting APC resistance, known as the activated partial thromboplastin time (APTT) assay. APC resistance is often implicated in thrombosis and certain pregnancy dysfunctions such as preeclampsia. The use of a fluorescence substrate offers simple, low-cost reactions with the potential to replace the existing clotting assay, which is time-consuming, costly, and complex.

The assay was developed by Morewood Molecular Sciences, Inc. (Pittsburgh, PA, USA) . The company has developed a system for performing highly multiplexed assays of enzyme activity on the surface of a chip and can conduct 5,000 assays in minutes on a one-by-three-inch surface using nanoliter quantities of reagent, without well plates or robotics.

The new assay may be used in conjunction with fluorescence or color assays to create highly multiplexed clotting tests, according to Morewood. "This aligns with Morewood's strategy to offer multiplexed assays for in vitro diagnostics,” said Christophe J-P. Sevrain, CEO of Morewood. "The complexity of coagulation pathways creates the need for systemic approaches in blood diagnostics, for which this technology is highly suited.”
